Abstract
Protocol from BLS Histology at University of Minnesota for processing snap frozen prospective surgically obtained tissue.
Guidelines
This protocol required prior approval by the users' local Institutional Animal Care and Use Committee (IACUC), Institutional Review Board (IRB) or equivalent ethics committee(s), depending on the source of the tissue.
Materials
PBS buffer
Cryovials
Liquid nitrogen bath

Troubleshooting
Before start
Prepare a liquid nitrogen bath large enough to hold a cryovial
Fresh Frozen Tissue
Tissue is received from surgical team and cut to a smaller size, ~1g
Tissue is placed in PBS buffer
When ready to freeze sample, remove tissue from PBS buffer and tap off excess buffer with a kimwipe
Tissue is placed in appropriately labeled cryovial
Snap freeze the cryovial in liquid nitrogen and store at -80°C
