Abstract
This protocol describes how to collect striatal punches from fresh frozen mouse brain tissue for HPLC analysis.
Troubleshooting
Striatal Punch Collection
Sacrifice mice with CO2, rapidly remove the brain, and freeze in superchilled isopentane on dry ice. Store brains at -80˚C until ready to collect tissue.
Embed brains in OCT, and mount on a cryostat. Trim until the striatum, then take punches from the dorsal striatum using a pipette tip cut to 1mm in diameter.
Store striatal punches at -80˚C until ready to ship for analysis. Ship on dry ice.
