Abstract
This protocol describes the setup and operation of the PX2+ Photometric Transmitter with an integrated fluorescence probe for real time fluorescence measurement and optional data logging. Two operating modes are described: use with the PX2+ Control Panel App for data logging and calibration, and standalone operation for rapid measurements without data collection. This protocol is intended for reproducible fluorescence measurements across users and experimental sessions.

Confirm that the fluorescence probe is securely connected.

Solution
Verify the USB connection and restart the application.
Before start
Ensure that all components are present and undamaged. Confirm that the PX2+ Control Panel App is available on the provided USB flash drive or already installed on the computer. Determine whether the experiment requires data logging or only real time fluorescence visualization.
Operation With PX2+ Control Panel App
Install the PX2+ Control Panel App on the computer using the provided USB flash drive.
Connect the integrated fluorescence probe to the PX2+ Photometric Transmitter.
Connect the PX2+ Photometric Transmitter to the computer using the USB cable.
Supply power to the PX2+ Photometric Transmitter only after the USB connection has been established.
Launch the PX2+ Control Panel App on the computer.
Verify that the device is recognized by the software interface.
Enable data logging by navigating to Tools, then Logging Setup, then Status, and selecting Enable Logging.
Select a file location for data storage. Fluorescence data will be recorded and saved as a CSV file.
Confirm that fluorescence signal values are updating in real time within the application interface.
Standalone Operation Without PX2+ Control Panel AppUntitled section
Ensure that the PX2+ Photometric Transmitter is not connected to a computer.
Connect the integrated fluorescence probe to the PX2+ Photometric Transmitter.
Supply power to the PX2+ Photometric Transmitter.
Observe real time fluorescence output via the device interface.
