Raman volume map of spectral peak intensities showing subcellular nucleic acid (red, wavenumber position 784 cm-1) and protein (cyan, wavenumber position 1002 cm-1; phenylalanine used as a protein proxy) distributions in a fully-hydrated, preserved Tetrahymena cell. Subcellular structures such as micro- and macro- nuclei (red) are resolved. Data set was acquired during a 13 hour Raman scan with a 633 nm He/Ne laser along a 3-D grid with a spatial resolution of 2 µm x 2 µm x 4 µm in XYZ dimensions, yielding 3472 individual spectra.
