Abstract
This protocol describes the placement of samples for XPCT and preparation of correlative histological analysis after the measurement.
Troubleshooting
XPCT-Sample Preparation
Selection of ROI in a FFPE tissue block and punch biopsy with 1 mm diameter to be placed in Kapton tube
perform XPCT measurement as described here:

Reembedding of FFPE tissue punch
perform at RT this delicate preparation step
hold Kapton tube with sample inside using fine forceps
push sample carefully out of the Kapton tube
Critical
prefill casting mold with warm paraffin and place it at RT
place sample with tissue facing the bottom in liquid paraffin during the curing process of paraffin
finally place cassette holder on top and place the new paraffin block on a cold plate
proceed to regular slicing using a microtome
