Abstract
Stocks are more permanent methods of storing bacteria than plates. These stocks can be recovered by pouring them into small volumes of BG-11 and growing under low light levels. As the culture grows, it can be passaged into higher volumes and appropriate antibiotics can be added.

Add your stock to Amount2 mL BG-11 medium in a test tube and grow it overnight.
Note
Do not add any antibiotics


Transfer this to a Amount50 mL flask and make up the culture to Amount10 mL in volume.

Once it reaches a lime green colour (#32CD32), add a fresh medium to make the culture up to Amount50 mL culture in a Amount150 mL

Further, make this culture up to Amount100 mL in a Amount250 mL flask once it returns to lime green.
