Zap them! We use 0.1 cm cuvettes from BioRad and zap the cells at 1.8 kV, 200 Ohms and 25 microFd, but preferred settings and cuvette types can vary from lab to lab. If it arcs, sparks or makes a weird popping noise: you have likely killed your cells *sad trumpet sound*. BUT: if you're feeling lucky, just proceed with the protocol and see if it worked, I have around a 30% success rate with arced samples, which is not terrible.
