Abstract
Standard protocol for the preparation of NGM medium for the maintenance and growth of C. elegans.
Based on C. elegans Methods and Applications, Third Edition - Gal Haspel
Materials
1. Buffer 1M KPO4 (6.0 pH):
a. 108.3 g KH2PO4
b. 35.6 g K2HPO4
c. Aforar a 1 Litro
d. Filtrar
e. Alicuotar y guardar a 4°C**
2. 5 mg/ml colesterol
a. Diluir en Etanol 100%
b. No filtrar**
c. Alicuotar y guardar a 4°C**
3. 10 mg/ml nistatina
a. Diluir en Etanol 70%
b. No filtrar**
c. Alicuotar y guardar a 4°C**
4. 1M MgSO4
a. Preparar y filtrar
b. Alicuotar y guardar a 4°C**
5. 1M CaCl2
a. Preparar y filtrar
b. Alicuotar y guardar a 4°C**
6. NaCl
7. Agar
8. Peptona
Troubleshooting
Métodos
Añadir 3 g de NaCl
25 g de Agar
2.5 g de Peptona
Añadir 975 ml de H2O destilada
Añadir 1 barra agitadora y AUTOCLAVAR**
Una vez autoclavado el medio: enfriar a baño maría a 55°C
Una vez a 55°C colocar en la placa de agitación a 55°C
Añadir 1 ml de 1M CaCl2
Añadir 1 ml 1M MgSO4
Añadir 1 ml de colesterol 5 mg/ml
Añadir 1.25 ml de nistatina 10 mg/ml
Añadir 25 ml 1M KPO4 buffer
Añadir Antibiótico en cantidad requerida.
Dosificar a conveniencia en platos pequeños o grandes
