Abstract
This protocol is suitable for preparing bacterial storage for -80 degree Celsius with glyceral as the solvent. It is usually performed after the overnight incubation of transformed bacteria that are ready for plasmid extraction.
Guidelines
The concentration of glycerol is optimized for around 17% in this protocol.

Make sure that all the steps should be operated in laminar flow.

Follow the "preparation" section of protocol.

Turn on the UV light of the laminar flow for Duration00:10:00 to Duration00:15:00 in order to sterilize it.

Add 300 μL of 50% glycerol into an eppendorf.
Add 600 μL of bacterial media prepared in a procedure like "Bacterial Culture for Plasmid Extraction" to an eppendorf.
Store the eppendorf under -80 degree Celsius.
