Preparation of samples for Jess
There is a detailed instruction included in the reagent pack from Bio Techne. The protocol was applied without changes.
Open EZ Standard pack. Reconstitute DTT (40 uL dH2O), Fluorescent 5X Master Mix (20 uL 10X Sample buffer and 20 uL DTT) and Biotinylated Ladder (20 ul dH20)
Dilute 10x sample buffer to 0.1x sample buffer in dH2O
Combine one-part fluorescent master mix with four parts of sample (final concentration of 1 μg/μl)
Boil samples and biotinylated ladder at 95 °C for 5 min
Load Separation Capillary Cartridges with 3 ul of sample per well and antibodies
Spin plate at 1000 x g for 5 min
Load Separation Capillary Cartridges with wash buffer, luminol-peroxide mix and RePlex reagent (if running replex protocol)
