Abstract
This is a protocol to explain how bacteria growing on NaOH crystals were grown and isolated.
Growing Bacteria From NaOH Crystals
A 0.5g sample of the sodium hydroxide crystals was suspended in 2mL of deionized water in a glass vial

Crystals were crushed in the vial to release any bacteria that may have been stuck in the crystals
DI water and sodium hydroxide crystals were mixed by hand for 5 minutes

Using a micropipette a 900 µL sample of the water and crystals was pipetted on the surface of LB agar at pH 7
Using the spread plate technique and a sterile spreader the liquid was spread around the plate
The medium was incubated aerobically at 35°C for 4 days to produce colonies
