Abstract
This protocol is for the preparation of 10% glycerol stocks of bacterial strains.
From a streaked plate of the bacteria, pick one colony and inoculate into a test tube containing Amount10 mL 1X LB
Grow the culture till it reaches an OD ~ 0.6
Into cryo-vials add Amount1.6 mL culture and Amount0.4 mL 50% autoclaved glycerol.

Note
This is for preparation of 10% glycerol stock. This can be changed according to the requirement.

Flash freeze the tubes in liquid Nitrogen and store it at Temperature-80 °C
