Abstract
This protocol details the DFP-induced mitophagy experiments.
Materials
Deferiprone (DFP) (379409, Sigma Aldrich)

Reagent3-Hydroxy-1,2-dimethyl-4(1H)-pyridoneMerck MilliporeSigma (Sigma-Aldrich)Catalog #379409

Troubleshooting
DFP-induced mitophagy
2d
To induce mitophagy,

  • Treat the cells for Duration24:00:00 with Concentration1 millimolar (mM) Deferiprone (DFP) (379409, Sigma Aldrich), an iron chelator that mimics hypoxic conditions through stabilization of the transcription factor HIF1α and subsequent upregulation of NIX and BNIP3.
1d
Freshly dissolve the compound in sterile dH2O on the day itself and boil for Duration00:01:00 at Temperature95 °C to dissolve the compound.
