Abstract
This is a collection of detailed step-by-step guides covering label-free Glycosaminoglycan (GAG) disaccharide analysis by HILIC-MS/MS of diverse biological sample types.

Methods are given for sample disruption, label-free GAG disaccharide generation, equimolar standard preparation, sample/standard vialling and HILIC-MS/MS data analysis and processing.

This analysis enables the relative quantification of GAG disaccharide amounts, total GAG amounts and disaccharide composition, from low amounts of diverse biological sample types.
