Abstract
The cellular morphology of M231 and MTS-M231 cells in monolayer culture was assessed using the Giemsa staining assay. Briefly, a total of 50,000 cells were seeded in each well of a 6-well plate and cultured for 3 days in complete medium. Subsequently, the cells were fixed with 70% ethanol for 10 minutes and stained with Giemsa dye (Beyotime) for 30 minutes. The nuclei were stained with a purplish-red color, while the cytoplasm exhibited a blue hue. The images were captured using a phase contrast microscope (Scope A1, Zeiss).
Troubleshooting
Cellular morphology assay of M231 by Giemsa staining
Seed a total of 50,000 cells in each well of a 6-well plate and culture for 3 days in complete medium.
Fix the cells with 70% ethanol for 10 minutes.
Stain the cells with Giemsa dye (Beyotime) for 30 minutes.
Observe that the nuclei are stained with a purplish-red color, while the cytoplasm exhibits a blue hue.
Capture images using a phase contrast microscope (Scope A1, Zeiss).
