Abstract
ATF5-inducible overexpressing human iPSCs
Troubleshooting
Transduce control human iPSCs with lentiviral vectors encoding doxycycline-inducible constructs for ATF5 overexpression (VectorBuilder ID: VB241111-1111fuf) or with an empty vector control (VectorBuilder ID: VB201122-1083bzq), both containing a neomycin resistance cassette.
After transduction, perform clonal selection with G418 at 150 μg/mL for two weeks.
Expand stable clones following selection with G418.
Use expanded stable clones for experiments without further antibiotic selection.
